Dormancy Breaking and In Vivo Growth of In Vitro Bulblets in Lilium Oriental Hybrid ¢¥ Casa Blanca ¢¥



Abstract
In vitro bulblets of Lilium Oriental Hybrid ¢¥Casa Blanca¢¥ were treated with cold temperature at 5¡É, and then planted in greenhouse with 2 weeks interval. Effects of in vitro sucrose concentrations and bulblet sizes on dormancy and in vivo growth of bulblets were investigated. GA©ý, GA_(4+7), and BA were also treated alone or in combination, for dormancy breaking of bulblets, and soil moisture for cold period were measured on the survival and growth of bulblets in vivo. More than 95% of leaf emergence was observed after 6 weeks at 5¡É cold treatment in the bulblets grown 3% of sucrose, 10 weeks in those from 6% of sucrose, and 12 weeks in those from 9% of sucrose. Thus, dormancy period of bulblets became longer as in vitro sucrose concentation increased. Any treatment effects of GA©ý, GA_(4+7), and BA alone or in combination were not found for bulblet dormancy breaking, but domancy breaking of bulblets was satisfied by only cold treatment. Cold period was extended as bulblet size increased. Leaf number and flower stalk emergence increased with extending cold period and increasing bulblet size. Bulblet survival was reduced, but the leaf emergence rate of survival bulblets was increased as cold period was extended. 50% of soil moisture during cold period was adequate for the survival and leaf emergence of bulblets.
